CPSF3 Promotes Pre-mRNA Splicing and Prevents CircRNA Cyclization in Hepatocellular Carcinoma
نویسندگان
چکیده
CircRNAs are crucial in tumorigenesis and metastasis, comprehensively downregulated hepatocellular carcinoma (HCC). Previous studies demonstrated that the back-splicing of circRNAs was closely related to 3′-end splicing. As a core executor cleavage, we hypothesized CPSF3 modulated circRNA circularization. Clinical data were analyzed establish prognostic correlations. Cytological experiments performed determine role HCC. A fluorescent reporter employed explore mechanism. The regulated by screened RNA-seq validated PCR, changes downstream pathways explored molecular experiments. Finally, safety efficacy inhibitor JTE-607 verified both vitro vivo. results showed highly expressed HCC cells, promoting their proliferation migration, high level predictive poor prognosis. mechanistic study revealed enhanced RNA thereby reducing circRNAs, increasing linear mRNAs. Furthermore, inhibition JET-607 suppressed cells. Our findings indicate increase promotes shift pre-mRNA from mRNA, leading uncontrolled cell proliferation. exerted therapeutic effect on blocking CPSF3.

TRAP150 activates pre-mRNA splicing and promotes nuclear mRNA degradation
TRAP150 has been identified as a subunit of the transcription regulatory complex TRAP/Mediator, and also a component of the spliceosome. The exact function of TRAP150, however, remains unclear. We recently identified TRAP150 by its association with the mRNA export factor TAP. TRAP150 contains an arginine/serine-rich domain and has sequence similarity with the cell death-promoting transcriptiona...
متن کاملAlternative mRNA splicing of liver intestine-cadherin in hepatocellular carcinoma.
PURPOSE To identify alternative splicing of the liver intestine-cadherin (LI-cadherin) gene in hepatocellular carcinoma (HCC) and correlate its aberrant expression with clinical outcomes. EXPERIMENTAL DESIGN Reverse transcription-PCR (RT-PCR) and quantitative real-time RT-PCR were used to examine alternative mRNA splicing and mRNA level of LI-cadherin in 50 paired tumor-peritumor tissues of 5...
متن کاملA splicing variant of Merlin promotes metastasis in hepatocellular carcinoma
Merlin, which is encoded by the tumour suppressor gene Nf2, plays a crucial role in tumorigenesis and metastasis. However, little is known about the functional importance of Merlin splicing forms. In this study, we show that Merlin is present at low levels in human hepatocellular carcinoma (HCC), particularly in metastatic tumours, where it is associated with a poor prognosis. Surprisingly, a s...
متن کاملSPF45-related splicing factor for phytochrome signaling promotes photomorphogenesis by regulating pre-mRNA splicing in Arabidopsis.
Light signals regulate plant growth and development by controlling a plethora of gene expression changes. Posttranscriptional regulation, especially pre-mRNA processing, is a key modulator of gene expression; however, the molecular mechanisms linking pre-mRNA processing and light signaling are not well understood. Here we report a protein related to the human splicing factor 45 (SPF45) named sp...
متن کاملRNAcatalyses nuclear pre-mRNA splicing
In nuclear pre-messenger RNA splicing, introns are excised by the spliceosome, a dynamic machine composed of both proteins and small nuclear RNAs (snRNAs). Over thirty years ago, after the discovery of self-splicing group II intron RNAs, the snRNAswere proposed to catalyse splicing. However, no definitive evidence for a role of either RNAor protein in catalysis by the spliceosome has been repor...
